DiFMUP is a fluorinated MUP, which is a commonly used fluorogenic alkaline phosphatase substrate. The reaction product of DiFMUP has excitation/emission maxima of ~358/450 nm, essentially identical to the spectral properties of MUP. Upon phosphatase-induced hydrolysis DiFMUP generates fluorinated 4-methylumbelliferyl fluorophore that has a lower pKa than that of the coumarin fluorophore generated by MUP, making DiFMUP a superior substrate for continuously assaying acid phosphatases at low pH than MUP. DiFMUP is also used for monitoring protein phosphatase activities.
