丙酮酸是一種酸性較弱的有機(jī)酸, 分子中同時(shí)具有羰基和羧基兩個(gè)官能團(tuán), 它除具有羧酸和酮的性質(zhì)外, 還具有α- 酮酸的性質(zhì), 是最簡單的α- 酮酸(屬于羰基酸)。丙酮酸是體內(nèi)產(chǎn)生的三碳酮酸, 它是糖酵解途徑的最終產(chǎn)物, 在細(xì)胞漿中還原成乳酸供能, 或進(jìn)入線粒體內(nèi)氧化生成乙酰CoA, 進(jìn)入三羧酸循環(huán), 被氧化成二氧化碳和水, 完成葡萄糖的有氧氧化供能過程。丙酮酸還可通過乙酰CoA 和三羧酸循環(huán)實(shí)現(xiàn)體內(nèi)糖、脂肪和氨基酸間的相互轉(zhuǎn)化, 因此, 丙酮酸在三大營養(yǎng)物質(zhì)的代謝聯(lián)系中起著重要的樞紐作用 [1] 。
抗氧化作用
有研究已表明, 丙酮酸能抑制鼠體內(nèi)氧自由基的氧化作用, 同時(shí)作為一種過氧化氫清除劑, 具有防止自由基損傷的作用, 已在心臟再灌注損傷和急性腎衰竭中證實(shí)具有保護(hù)機(jī)體抗功能性損傷。丙酮酸可通過兩種機(jī)制起到抗氧化作用: 其一, 作為一種α- 酮酸, 丙酮酸可直接通過非酶促的去碳酸基反應(yīng)抑制過氧化氫; 其二,補(bǔ)充丙酮酸可增強(qiáng)檸檬酸循環(huán), 檸檬酸產(chǎn)生增多后, 抑制磷酸果糖激酶, 從而進(jìn)入磷酸戊糖旁路, 產(chǎn)生還原型輔酶Ⅱ(NADPH) , 從而間接地增加谷胱甘肽(GSH) 抗氧化系統(tǒng)的能力。丙酮酸還可增加輔酶Ⅰ/ 還原型輔酶Ⅰ(NAD+/NADH) 的比值, 促進(jìn)三羧酸循環(huán)反應(yīng)。 [2]
